Evaluation of The Effects of Desipramine,Venlafaxine And Bupropion on Depressionand Anxiety in Mice By Forced Swimming Testand Elevated Plus Maze Test

Öz:
Aim This study aimed to investigate the effects of desipramine, venlafaxine and bupropion on depression and anxiety in mice. We used a classic tricyclic antidepressant, desipramine; a serotonin-norepinephrine reuptake inhibitor, venlafaxine; and a second-generation antidepressant agent that blocks the central reuptake of noradrenaline and dopamine, bupropion. ( Sakarya Med J 2018, 8(4):820-829 )Material and MethodsImipramine (30 mg/kg), desipramine (7.5 mg/kg and 15 mg/kg), venlafaxine (4 mg/kg and 8 mg/kg), and bupropion (20 mg/kg and 40 mg/kg) signifi cantly reduced immobility time in the FST compared to vehicle in a dose-dependent manner. Additionally, bupropion (40 mg/kg) was signifi cantly more effective than imipramine.Results Diazepam (2 mg/kg), desipramine (7.5 mg/kg and 15 mg/kg), venlafaxine (4 mg/kg and 8 mg/kg), and bupropion (20 mg/kg and 40 mg/kg) signifi cantly increased the time spent in the open arms of the EPM compared to vehicle in a dose-dependent manner. Diazepam (2 mg/kg), desipramine (7.5 mg/kg and 15 mg/kg), venlafaxine (8 mg/kg) and bupropion (20 mg/kg and 40 mg/kg) signifi cantly increased the number of entries into the open arms compared to the vehicle-treated group. Additionally, bupropion (40 mg/kg) was signifi cantly more effective than diazepam.Conclusion Desipramine, venlafaxine and bupropion (20 mg/kg) exerted signifi cant antidepressant-like effects that were not signifi cantly different than the effects of imipramine. Additionally, bupropion (40 mg/kg) exerted signifi cant antidepressant-like effects and was more effective than imipramine. Similarly, desipramine, venlafaxine and bupropion (20 mg/kg) exerted signifi cant anxiolytic-like effects that were not signifi cantly different than the effects of diazepam. Additionally, bupropion (40 mg/kg) exerted signifi cant anxiolytic-like effects and was more effective than diazepam.

Öz:
Amaç Bu çalışmada, farelerde desipramin, venlafaksin ve bupropiyonun depresyon ve anksiyete üzerine etkilerini araştırmayı amaçladık. ( SakaryaTıp Dergisi 2018, 8(4):820-829 ). Gereç veYöntem Klasik trisiklik antidepresan, desipramin, serotonin-norepinefrin geri alım inhibitörü venlafaksin ve ikinci jenerasyon antidepresan bir ajanolan noradrenalin ve dopamin gerialım inhibitörü bupropionun farelerde zorunlu yüzme testi (FST) ve yükseltilmiş artı labirent testi (EPM)üzerindeki etkilerini araştırmayı amaçladık. Bulgular Imipramin (30 mg/kg), desipramin (7,5 mg/kg ve 15 mg/kg), venlafaksin (4 mg/kg ve 8 mg/kg) ve bupropion (20 mg/kg ve 40 mg/ kg) FST’de hareketsizlik zamanını önemli ölçüde doza bağlı bir şekilde azalttı. Ayrıca bupropion (40 mg/kg) da imipramine göre anlamlıderecede etkiliydi.Diazepam (2 mg/kg), desipramin (7.5 mg/kg ve 15 mg/kg), venlafaksin (4 mg/kg ve 8 mg/kg) ve bupropion (20 mg/kg ve 40 mg/kg)EPM’de açık kollarda kalış süresini önemli ölçüde doza bağlı arttırdı. Diazepam (2 mg / kg), desipramin (7,5 mg/kg ve 15 mg/kg), venlafak sin (8 mg/kg) ve bupropion (20 mg/kg ve 40 mg/kg) açık kola giriş sayısını da artırdı. Ayrıca bupropion (40 mg/kg) diazepamdan anlamlıderecede daha etkiliydi. Sonuç Desipramin, venlafaksin ve bupropion (20 mg/kg), imipraminin etkilerinden önemli ölçüde farklı olmayan antidepresan benzeri etkilereneden olmuştur. Bunun yanında bupropion (40 mg/kg) antidepresan benzeri etki göstermiştir ve bu etkiler imipraminden daha güçlüdür.Benzer şekilde, desipramin, venlafaksin ve bupropion (20 mg/kg), diazepamın etkilerinden önemli ölçüde farklı olmayan anlamlı anksiyolitikbenzeri etkiler göstermişlerdir. Ayrıca bupropion (40 mg / kg) anlamlı anksiyolitik benzeri etkiler göstermiş ve bu etkiler diazepamdan dahagüçlüdür.
