Prothrombotic Gene Polymorphisms in Young Patients with Cerebrovascular Accident

Öz:
Amaç: Serebrovasküler hastalıklar, insidansı yaşla birlikte artan, genetik veçevresel faktörlerin etkilediği kompleks multifaktöryel bozukluklardır. Serebrovasküler hastalıklara ait risk faktörleri yaş, cinsiyet, ırk, hipertansiyon, diabetes mellitus, hiperkolesterolemi vb. olsa da 45 yaş altındaki genç serebrovasküler hastalarda genetik faktörler etiyolojide rol oynayabilmektedir. Buretrospektif çalışmada serebrovasküler olay geçirmiş genç erişkinlerde hastalığın oluşmasında etkisi olabileceği düşünülen protrombotik gen polimorfizmleri araştırılmıştır. Gereç ve Yöntem: 45 yaş altındaki genç serebrovasküler olay geçiren 43 hastada Methylenetetrahydrofolate reductase (MTHFR)geni C677T ve A1298C polimorfizmleri ve Protrombin (Faktör II) G20210A,Faktör V Leiden G1691A protrombotik gen polimorfizmleri değerlendirildi.Bulgular: Serebrovasküler olay geçirmiş 43 genç erişkinde, MTHFR C677Tpolimorfizmi sıklığı %46.51 heterozigot, %9.3 homozigot, MTHFR A1298Cpolimorfizm sıklığı %39.47 heterozigot, %26.31 homozigot, Protrombin polimorfizm sıklığı %2.3 heterozigot, %2.3 homozigot, Faktör V Leiden polimorfizm sıklığı ise %9.3 heterozigot, olarak bulundu. Tartışma: Yapılan değerlendirme sonucu MTHFR geni C677T ve A1298C polimorfizmlerinin serebrovasküler olaylarda risk faktörü oluşturabileceği düşünülmüştür. Bu polimorfizmlerle birlikte sigara kullanımı, hipertansiyon ve aile öyküsünün bulunmasınında mevcut riski artırdığı gözlenmiştir.

Serebrovasküler Olay Geçiren Genç Hastalarda Protrombotik Gen Polimorfizmleri

Öz:
Aim: Cerebrovascular diseases are complex multifactorial disorders showingan increased incidence with increasing age and affected by genetic and environmental factors. Although risk factors for cerebrovascular diseases includeage, sex, lineage, hypertension, diabetes mellitus, hypercholesterolemia;in young cerebrovascular patients below age 45, genetic factors may alsocontribute to the etiology. In this retrospective study, prothrombotic genepolymorphisms which are thought to be related with formation of disease inyoung adults with cerebrovascular accident (CVA) were investigated. Material and Method: In the current study, Methylenetetrahydropholate Reductase (MTHFR) C677T and A129C; Prothrombin (Factor II) G20210A; FactorV Leiden G1691A prothrombotic gene polymorphisms were evaluated for43 young patients under the age of 45 with cerebrovascular accident his- tory. Result: For 43 young patients with cerebrovascular incident history, thefrequency of following polymorphisms were determined as follows; MTHFRC677T polymorphism heterozygous frequency is 46.1%, homozygous fre- quency is 9.3%; MTHFR A1298C polymorphism heterozygous frequency is39.47%, homozygous frequency is 26.31%; Prothrombin polymorphism heterozygous and homozygous frequency is 2.3%; FactorV Leiden polymorphism heterozygous frequency is 9.3%. Discussion: After evaluation the experimental results, we believe that MTHFR gene C677T and A1298C polymorphismsmight be risk factors in CVAs. It was observed that cigarette usage, hypertension and existence of family story in addition to these polymorphismsincrease the available risk.
