Flower type affects the timing of embryo development in native Primula vulgaris

Öz:
In this study, pin and thrum flowered wild primrose (Primula vulgaris) populations, which are naturally distributed in black sea region in Türkiye, were cultivated and their pollen viability, pollen tube growth and embryo development stages were examined. As a result of the study, pollen viability in pollen of the pin flower type was 83.10%, while the in vitro germination rate was 69.43%. The viability and germination rates of pollen of thrum flower type were 84.91% and 67.92%, respectively. As a result of the crush preparation examinations, it was observed that the pollen tube of both types started to germinate on the stigma on the 1st day. On the 4th day, it was determined that the pollen tubes penetrated to ovule. When the embryo development stages were examined, it was observed that zygote formation occurred on the 6th day after pollination in the pin flower type, and on the 20th day, the embryo was in the heart stage and on the 40th day, it was seen that it formed a mature embryo that reached the cotyledon stage. It was determined that the endosperm in the thrum type started to develop faster than the pin type and started to form on the 6th and 7th days. On the 10th day, it was determined that endosperm tissue was formed. In thrum flower type, it was determined that the proembryo started to form on the 10th day, but embryo formation could only be seen on the 30th day. In the ovarian examinations performed on the 40th day, it was determined that some embryos were in the cotyledon stage while some were still in the globular or cardiac stage. It was determined that no embryos were found in some ovules.
