Protective Effect of Ganoderma Lucidum Extract in Contrast Agent Induced Renal Toxicity

Objective: In this study, it was aimed to investigate the protec tive effect of Ganoderma lucidum extract (GANO) in kidney toxicity developed with a contrast agent in rats who devel oped acute kidney injury with thirst and furosemide (diuretic) administration. Methods: The rats were randomly grouped as follows: Sham, GANO200, RK (radiocontrast), RK+ISO, RK+GANO100, and RK+GANO200. Before administration of the contrast agent, rats were applied thirst and furosemide to develop acute kid ney injury. GANO was given to the determined groups 1 hour before the contrast agent application. Afterward, RK (15 mL/kg) was administered from the tail vein of the rats. The kidney tissues were examined histopathologically and biochemically. Results: In the Sham and GANO200 groups, the pathological finding was not observed in the glomerulus, proximal, and distal tubules. While shrinkage was observed in the renal corpuscu lar structures in the RK group, epithelial vacuolization was also seen in the proximal and distal tubules. Inflammatory cells were observed around the glomerular and distal tubules. The find ings of the RK+GANO200 were found to be similar to the sham group. It was determined that the MDA level increased in the RK group, while the GSH level decreased. With GANO applica tion, a decrease in MDA level and an increase in GSH level were observed. It was determined that serum creatinine and BUN levels increased in the radiocontrast agent applied groups and decreased after GANO application. Conclusion: After contrast medium injury, G. lucidum extract application may play a protective role in renal tubules, especially in cases of acute kidney injury.

Kontrast Madde ile İndüklenmiş Renal Toksisitede Ganoderma Lucidum Ekstraktının Koruyucu Etkisi

Öz:
Amaç: Bu çalışmada, susuzluk ve diüretik ilaç (Furosemide) uygulaması ile akut böbrek hasarı gelişen sıçanlarda, kontrast madde ile geliştirilen böbrek toksisitesi üzerine Ganoderma lucidum ekstraktının (GANO) koruyucu etkisinin araştırılması amaçlanmıştır. Yöntem: Deneye başlamadan önce sıçanlar rastgele aşağıdaki şekilde gruplandırıldı; SHAM, GANO200, RK (Radyokontrast), RK+İZO (RK+İzotonik), RK+GANO100 ve RK+GANO200. Kontrast madde uygulanmasından önce, sıçanlarda akut böb rek hasarı geliştirmek için susuzluk ve Furosemid uygulandı. Belirlenen gruplara kontrast madde uygulamasından 1 saat önce Ganoderma lucidium ekstresi verildi. Daha sonra sıçanların kuy ruk damarından yavaş yavaş Radyokontrast madde (15 mL/kg) uygulaması yapıldı. 24 saat sonra deney sonlandırılarak böbrek dokuları, histopatolojik ve biyokimyasal olarak incelendi. Bulgular: SHAM ve GANO200 gruplarında glomerül, proksimal ve distal tübüllerde patolojik bulgu izlenmedi. RK grubunda, renal korpüsküler yapılarda büzülme görülürken proksimal ve distal tübüllerde epitelyal vakuolizasyon görüldü. Ayrıca glome rül ve distal tübüllerin çevresinde inflamatuar hücreler gözlendi. RK+GANO200 grubunda, RK grubuna göre proksimal ve distal tübüllerde epitelyal vakuolizasyonun azaldığı ve bu grubun Sham grubuna benzer olduğu görüldü. Biyokimyasal parametreler değerlendirildiğinde MDA seviyesinin RK grubunda arttığı, GSH düzeyinin ise önemli ölçüde azaldığı belirlendi. GANO uygulaması ile MDA seviyesinde azalma, GSH seviyesinde ise artış izlendi. Serum kreatinin ve BUN (kan üre azotu) düzeylerinin radyokont rast madde uygulanan gruplarda arttığı, GANO uygulamasından sonra ise azaldığı belirlendi. Sonuç: Kontrast madde hasarından sonra, Ganoderma lucidium ekstresi uygulaması, özellikle akut böbrek hasarı durumlarında böbrek tübüllerinde koruyucu rol oynayabilir.
