In vitro seed germination study in endemic plant Onosma discedens

Öz:
Onosma discedens (Kemaliye Emzikotu), Erzincan yöresinde (Türkiye) oldukça dar yayılış alanına sahip nadir endemik bitki türlerindendir. Bu çalışmada Onosma discedens türünde farklı NaCl, HCl, KNO3, GA3 konsantrasyonları ve sıcak/soğuk/mekanik stratifikasyon uygulamalarının çimlenme başarısına etkilerinin incelenmesi amaçlanmıştır. Tohumlar 100 ve 200 ?M konsantrasyonlarda NaCl, HCl, KNO3, GA3 solusyonları kullanılarak ya da sıcak/soğuk/mekanik stratifikasyon uygulamalarının ardından 23/18°C'de 8 saat aydınlık/16 saat karanlık ortamda çimlendirilmiştir. Kontrol grubundan (% 7), daha yüksek çimlenme gerçekleşen uygulamalar; KNO3-100 µM (% 12), GA3-100 µM (% 25), GA3-200 µM (% 35), HCI-100 µM (% 9). Çimlendirme hızı katsayısının ise en yüksek potasyum nitratın uygulamalarında (9,5) olduğu bulunmuştur. Bu çalışma sonucunda, elde edilen veriler türün ex-situ korunması amacıyla kullanılabilinir.

Endemik Onosma discedens tohumlarında İn vitro çimlendirme çalışmaları

Öz:
Onosma discedens is endemic species having considerable narrow distribution in Erzincan (Turkey) Region. In this study, we investigated the germination behavior of Onosma discedens with different concentrations of NaCI, HCl, KNO3, GA3 (100 and 200 ?M) or hot-cold stratification/mechanical scarification. Seeds under the light and dark treatments were exposed to a photoperiod of 8 h light/16 h dark hours with a 23/18 °C thermoperiod. Germination rates increased with KNO3-100 µM (% 12), GA3-100 µM (% 25), GA3-200 µM (% 35), HCI-100 µM (% 9) treatments when compared to the control. The highest speed of germination index was obtained at concentrations of 100-200 µM KNO3. Overall, our results will provide valuable data for ex situ conservation of endemic Onosma discedens.
