Determination of genetic and morphologic diversity of Cyperus difformis L. (Smallflower umbrella sedge) in rice growing areas

Öz:
Cyperus difformis L. (Smallflower umbrella sedge) is one of the important weedspecies in the rice production areas worldwide. To investigate the effect ofthe morphological and genetic diversity to the existing herbicide resistance inTurkey, it was studied with 50 populations to represent the rice growing areas.To determine the morphological diversity, 13 different characteristics wererecorded. The results of the analysis showed a morphologically significantdifference between the populations. The genetic variation among populationswas determined by Random Amplified Polymorphic DNA (RAPD) analysisusing 17 oligonucleotide primers. According to the dendrogram informationgenerated using Average Linkage, populations were classified into two maingroups at 0.25 genetic distance. The morphological and molecular analysesrevealed differences in terms of several quantitative characteristics amongthe populations examined. Similarities were also found among differentC. difformis population grown in different regions in terms of their morphologicalcharacteristics, the genetic diversity was found to be higher. This indicates ahigh potential at the point of gene transfer. We have concluded that the transferof seeds between regions and the resistance developed by weed againstherbicides and adaptation of species to the geographical area may be causeto these results. In this situation, it is necessary to focus on the control ofC. difformis seed movement, rice seed trafficking and the use of herbicide usesas well as in the field management practices such as or cultural methods. Itshould not be forgotten that the measures to be taken at this point are importantat the point of integrated management strategies.

Çeltik ekim alanlarında sorun olan Cyperus difformis L. (Kız otu)’in genetik ve morfolojik çeşitliliğinin belirlenmesi

Öz:
Cyperus difformis L. (Kız otu) dünya genelinde çeltik üretim alanlarında sorun olan önemli yabancı ot türlerinden biridir. Türün morfolojik ve genetik çeşitliliğinin saptanmasıyla, ülkemizdeki mevcut dayanıklılık olgusuna etkisinin belirlenmesi hedeflenen çalışma kapsamında, çeltik ekim alanlarını temsil edecek şekilde 50 popülasyon ile çalışılmıştır. Morfolojik farklılığı saptamak için 13 farklı parametre incelenmiştir. Analiz sonuçları popülasyonlar arasında morfolojik olarak belirgin farklılığı göstermiştir. Popülasyonlar arasındaki genetik varyasyon 17 oligonükleotid primeri kullanılarak Rasgele Çoğaltılmış Polimorfik DNA (RAPD) analiziyle belirlenmiştir. Average Linkage kullanılarak oluşturulan dendogram bilgilerine göre 0.25’lik genetik uzaklıkta çalışılan popülasyonların iki ana gruba ayrıldığı görülmüştür. Yapılan morfolojik ve moleküler analizlerde incelenen popülasyonlar arasında bazı kantitatif karakterler noktasında farklılıklar tespit edilmiştir. Farklı bölgelerde yetişen popülasyonlar arasında morfolojik bakımdan göz ardı edilmeyecek düzeylerde benzerlikler saptanırken, genetik olarak çeşitlilik daha yüksek bulunmuştur. Bu durum gen taşınması noktasında yüksek bir potansiyeli işaret etmektedir. Saptanan olgunun coğrafi alanlara adaptasyon, insanlar ve aletler tarafından tohumların bölgeler arası taşınması ve yabancı ot mücadele yöntemleri içerisinde özellikle kullanılan herbisitlere karşı yabancı ot tarafından geliştirilen dayanıklılıktan kaynaklanabileceği kanısına varılmıştır. Bu bağlamda C. difformis tohum hareketinin azaltılması, çeltik tohumluk trafiğinin ve herbisit kullanımının iyi denetlenmesinin yanısıra, kültürel yöntemler gibi alan yönetimi uygulamalarına odaklanılması gerekmektedir. Bu noktada alınacak önlemlerin entegre mücadele stratejileri açısından önem arz ettiği unutulmamalıdır.
