TY - JOUR TI - EVALUATION OF CYTOXICITY OF QMIX, ETHYLENE DIAMINTETRAACETIC ACID AND CHLORHEXIDINE ON HUMAN OSTEOBLAST CELL LINE AB - Objectives: In this study, the time-dependent toxiceffects of QMix ™, ethylene ediaminetetraacetic acid andchlorhexidine irrigation solutions on human osteoblastcells were as evaluated.Methods and Materials: Human osteoblast cells weregrown as monolayer cultures at 37ºC in an atmosphere of5% CO2 in air and 100% relative humidity. Cells wereexposed to ethylene diaminetetraacetic acid (EDTA),chlorhexidine (CHX) and QMix™ for 4 hours and 24hours. Cell viability was assessed by a 2,3-bis(2-methoxy-4-nitro-5-sulfophenyl)-5-[(phenylamino)carbonyl]-2H-tetrazolium hydroxide kit(XTT) assay. The differences in the mean viability ofhuman osteoblast cells were evaluated statistically.Results: There was a statistically significant differencebetween the mean percentage of viable cells in the testsolutions and control group, both after 4 hour (p˂0.001)and 24 hour exposure (p=0.004). The mean percentage ofviable cells decreased statistically significantly with theincrease in the time of exposure in the EDTA, CHX andQMixTM groups (p˂0.05). After 4 hours’ exposure, theEDTA and QMix showed statistically a less toxic effectthan did CHX (p˂0.05). There was no statisticallysignificant difference between the toxicity of theirrigation solutions after 24 hours’ exposure (p>0.05).Conclusion: All irrigation solutions tested showedvarious toxic effects on the human osteoblast cell line.The increase in exposure time also increased the toxicityof irrigation solutions on the human osteoblast cell line.
