Overektomi uygulanan sıçanlarda uterus dokuları üzerinde astaksantinin etkisi: histopatolojik çalışma

Öz:
Amaç: Güçlü bir antioksidan olarak bilinen astaksantin (AST), inflamasyona karşı yararlı etkilere sahiptir. Bu çalışmada, overektomili sıçanlarda astaksantinin uterus dokuları üzerindeki etkisi araştırılmıştır. Gereç ve Yöntem: Sprague-Dawley sıçanlar (n = 18); rastgele üç gruba ayrıldı (n = 6): sağlıklı kontrol, overektomi (OVT) ve overektomi+ astaksantin (OVT + AST). AST (1 mg / ml), overektomiden dört hafta önce başlanarak günlük 50 ml su ile deney grubuna oral yoldan verildi. AST tedavisi overektomi sonrası dört hafta daha sürdürüldü. Sekizinci haftanın sonunda, hayvanlar servikal dislokasyon ile ötenazi edildi. Ötenazi sonrası uterus formalin ile sabitlendi ve Hematoksilin-Eozin boyaması ile histolojik açıdan incelendi. Bulgular: OVT ile ortaya çıkan uterus epitelinde apiko-bazal yükseklikteki azalma ve hücresel bozulma, uterus bezlerindeki dejenerasyon ve stromadaki inflamasyon belirtileri, AST uygulanan sıçanlarda kontrole yakın düzelme göstermiştir. Sonuç: Tuba uterina, uterus ve vajinada menopoz sonrası ortaya çıkan değişiklikler menopozun klinik bulgularıyla yakından ilişkilidir. Astaksantinin bu dokular üzerindeki olumlu etkisinin mekanizmasının araştırılması gerekmektedir.

Effect of astaxanthin on uterus tissues in ovariectomized rats: a histopathological study

Öz:
Objective: Astaxanthin (AST), known as a powerful antioxidant, has beneficial effects against inflammation. In this study, the effect of astaxanthin on uterine tissues in ovariectomized rats was investigated. Material and Methods: Sprague-Dawley rats (n = 18); randomly divided into three groups (n = 6): healthy control, ovariectomy (OVT) and ovariectomy + astaxanthin (OVT + AST). AST (1 mg/ml) was given orally to the experiment group with 50 ml of water daily starting four weeks prior to ovariectomy. AST treatment was continued for another four weeks after ovariectomy. At the end of the 8th week, animals were euthanized by cervical dislocation. The uterus removed after euthanasia was fixed with formalin and examined with Hematoxylin & Eosin staining. Results: The symptoms of decrease in epithelial apico-basal thickness and cellular deterioration in the uterine epithelium caused by OVT, degeneration in the uterine glands, and inflammation in the stroma showed improvement in AST treated rats. Conclusion: The changes occurring in the tuba uterina, uterus, and vagina after atrophic menopause are closely related to the clinical manifestations of menopause. The mechanism of the positive effect of astaxanthin on these tissues should be investigated.
