Biodegradation of heptachlor by Phanerochaete chrysosporium ME 446: The toxic effects of heptachlor and its metabolites on mice

Beyaz çürükçül funguslar içinde yer alan Phanerochaete chrysosporium ME446'un toksik, mutajenic ve karsinojenik özelliklere sahip olan çok sayıda organik bileşiği yıkma özelliğinden yola çıkarak; yine aynı özelliklere sahip olan ve organik klorlu insektisitler içinde yer alan heptaklorun (1, 4, 5, 6, 7, 8, 8-heptaklor -3a, 4, 7, 7a-tetrahidro- 4, 7- methanoin) söz konusu fungus ile yıkılmasında etkili fizyolojik koşullar saptanmıştır. Ayrıca heptaklor ve yıkımı sonucu oluşan metabolitlerinin fareler üzerinde oluşturabileceği toksik etkiler araştırılmıştır.Fizyolojik koşulların saptanmasına ilişkin çalışmalarda-, heptaklor yıkımında optimum pH S, inkübasyon süresi 20 gün, inkübasyon sıcaklığı 30°C -35°C ve heptaklor konsantrasyonu 50 mM olarak saptanmıştır. Optimum fizyolojik koşullar uygulandığında heptaklor için maksimum yıkım oranı %97.30 bulunmuştur. Toksikolojik değerlendirmeler ile ilgili çalışlarda öncelikle heptaklorun LD50 dozu saptanmış olup, farelerin kanındaki şekilli elemanlar üzerine etkisi incelenmiştir. Heptaklor için LD50 dozu 97.50: mg/kg bulunmuştur. Farelerde %50 ölüme neden olan konsantrasyondaki heptaklorun uygun fizyolojik koşullarda yıkımı ile oluşan metabolitler farelere uygulandığında, farelerin hiçbirinde ölüme yol açmadığı görülmüştür. Ayrıca heptaklor farelere doğrudan uygulandığında kanın şekilli elemanları üzerine niceliksel bir etki gösterirken, metabolitlerin farklı bir etki göstermediği gözlenmiştir.

Heptaklor'un Phanerochaete chrysosporium ME 446 ile biyolojik yıkımı: Heptaklor ve metabolitlerinin fareler üzerine toksik etkisi

On the basis of degradation properties of Phanerochaete chroysosporium ME 446 found in white-rot fungi in respect of many organic compounds with toxic, mutagenic and carcinogenic characteristics, the physiological conditions effective for the degradation of heptachlor (1, 4, 5, 6, 7, 8, 8-heptachloro-3a, 4, 7, 7a-tetrahydro-4, 7-methanoindene), an organic chlorine used in insecticides Phanerochaete chrysosporium ME were determined. The potential toxic effects of heptachlor and metabolites on mice were also investigated. For the degradation of heptachlor, the optimum pH was found to be 5, incubation period 20 days, and incubation temperature, 30°C-35°C, while the optimum concentrations was 50 $mu$ M. The maximum heptachlor degradation rate under optimum physiological conditions was found to be 97.30%. The LD50 dose of heptachlor was determined, and its effects on cells in the blood of mice were investigated. The LD50 dose was found to be 97.50 mg/kg for heptachlor. Although this dose normally produces a death rate of 50% in mice, the resultant metabolites obtained through Phanerocheate chrysosporium ME 446 treatment caused no deaths. While direct application of heptachlor produced quantitive effects in the blood cells, no significant effects of heptachlor metabolites could be detected.
