Niğde Tepecik-Çiftlik ve Konya Çatalhöyük kazılarına ait insan kemiklerinden antik DNA eldesi ve genom dizilemesi yoluyla Orta Anadolu Neolitik Çağ insan popülasyonlarının karakterizasyonu

Öz:
Sunulan çalışmanın ana amacı Neolitik Dönem Orta Anadolu insan popülasyonlarının demografik tarihlerinin araştırılması ve bu yolla Neolitik Dönüşüm sürecinin dinamiklerinin anlaşılmasıdır. Çalışmanın ilk aşamasında üç Orta Anadolu Neolitik yerleşiminden çıkarılmış 70'i aşkın insan kemik ve diş örneğinden ODTÜ Antik DNA Laboratuvarı'nda DNA saflaştırılmış, bunlardan yeni nesil dizileme kütüphaneleri oluşturulmuş, ön dizilemeler sonucunda insan DNA'sı görece yüksek ve kontaminasyon göstermeyen örnekler seçilerek hibridizasyonla tüm genom hibridizasyonla yakalama işlemine tabi tutulmuş ve derinlemesine dizilenmiştir. Konya Boncuklu Höyük'ten (M.Ö. 8300-7500) dört ve Niğde Tepecik-Çiftlik'ten (M.Ö. 7500-5800) beş bireye ait toplam dokuz genom popülasyon genetiği analizine izin verecek derinlikte dizilenmiştir. Çalışmanın ikinci aşamasında dizilenen genom verisi erken Holosen'e ait yayımlanmış antik genom verileriyle karşılaştırılmiş ve bir dizi bulgu elde edilmiştir: (1) Orta Anadolu'da Neolitik Dönem'de yaşamış toplumlar, Avrupa'ya M.Ö. 7000 sonrasında doğudan göç eden ve bu kıtaya tarımı getiren ilk çiftçi gruplarla yakın akrabadır. (2) Erken ve geç dönem Anadolu Neolitik toplumlarını karşılaştırınca, zaman içinde genetik çeşitlilik artmış gözükmektedir. Bu da Neolitik süresince nüfus hareketlerinin ve gruplar arasında karışımın arttığına işaret etmektedir. Çalışmanın üçüncü safhasında ise projede üretilen aynı veriler, 2016'da başka gruplarca yayımlanan yeni antik DNA verileriyle karşılaştırılmıştır. Bu analiz (1) Orta Anadolu'nun Boncuklu tarafından temsil edilen erken Neolitik popülasyonlarının Levant'tan veya İran'dan gelen Neolitik göçmenler olmadığını, dolayısıyla muhtemelen yerli avcı toplayıcıların torunları olduklarını göstermiştir. (2) Ege'de M.Ö. 7000'den sonra ortaya çıkan ilk çiftçilerin de Egeli avcı toplayıcı grupların torunları olabilecekleri, yani kültürel etkileşim yoluyla Neolitikleşmiş olabilecekleri ihtimalini ortaya koymuştur. Çalışmada ayrıca tarafımıza ulaştırılan 30 adet Konya Çatalhöyük (M.Ö. 71005900) insan örneğin üzerinde farklı DNA özütleme ve kütüphane hazırlama yöntemleri denenmiş ancak bu örneklerde insan DNA'sı içeriğinin düşük olduğu tespit edilmiştir. Sonuçlarımız, Anadolu'da Neolitik Döneme ait örneklerde tüm genom analizinin mümkün olduğunu, ancak yerleşimler arasında DNA korunma oranı açısından ciddi farklar bulunduğunu göstermiştir. Popülasyon genetiği analizleri ise Neolitik Dönüşüm sürecinde hem Orta Anadolu hem de Ege'de yerel avcı toplayıcıların önemli rolü olabileceğine işaret etmektedir.

Öz:
The main purpose of this study was the characterization of the demographic history of Neolithic Period Central Anatolian populations and thereby achieving a fuller understanding of the Neolithic Transition. At the first stage we isolated DNA from more than 70 bone and tooth samples from three Central Anatolian Neolithic sites at the METU Ancient DNA Laboratory, and prepared next generation sequencing libraries from these for prescreening. Four Konya Boncuklu Höyük (8300-7500 cal BCE) and five Niğde Tepecik-Çiftlik (7500-5800 cal BCE) individuals’ samples contained relatively high amounts of human DNA and no indication of contamination, and were chosen for whole genome capture by hybridization and deep sequencing reaching levels sufficient for population genetics analysis. At the second stage we compared this genome data with published early Holocene genomes, yielding the following results: (1) The central Anatolian Neolithic populations were closely related to the first farmer populations in Europe who migrated there after 7000 cal BCE, bringing farming to the continent. (2) Comparing early and late Neolithic populations in Anatolia, we discovered that genetic diversity increased in time, implying that during the Neolithic Transition population movements and admixture events increased in frequency. At the third stage the same data was compared with ancient genome datasets published in 2016 by other groups. These analyses showed that (1) Central Anatolian early Neolithic populations (at least Boncuklu) were not migrants from the Levant or from Iran, and were thus most likely the descendants of local hunter gatherer groups. (2) We found that the early farmer populations that emerged in the Aegean post-7000 BCE could have been the descendants of local Aegaen hunter gatherer groups, and thus may have became Neolithized through cultural interactions with their eastern neighbors. Finally, we applied different DNA extraction and library preparation techniques on a total of 30 Konya Çatalhöyük (7100-5900 cal BCE) samples, but found that these generally contained low human DNA proportions. Our results document the possibility of obtaining ancient DNA from early Holocene samples in Anatolia, despite significant variation among sites in DNA preservation. Meanwhile, our population genetic analyses suggest a role for local hunter gatherers both in central Anatolia and in the Aegean in the Neolithization of these regions.
