Endodontik İnfeksiyonların Yeni Nesil Sekanslama Metoduyla Analizi

Öz:
Bu çalışmanın amacı primer ve sekonder/persistan endodontik infeksiyonlarda kanal içindeki mikrobiyotanın yeni nesil sekanslama tekniği ile mikroorganizma topluluk profilini araştırmaktır. Çalışmaya 20 primer endodontik ve 20 sekonder/persistan endodontik infeksiyonlu 40 diş dahil edildi. Dişler atravmatik şekilde çekildikten sonra kriyojenik pulverizasyonla toz haline getirildi. Toz haline getirilen örneklerden mikrobiyal DNA ekstrakte edildi. 16S rRNA geninin V4 bölgesi amplifiye edildi ve elde edilen amplikonlar 454 GS FLX cihazı kullanılarak pirosekanslama işlemine tabi tutuldu. 40 örnekten toplam 2.606.128 dizi okuması, örnek başına 64.700 adet kaliteli okuma gerçekleştirildi. Çalışmada toplam 14 filum, 29 sınıf, 47 takım, 85 aile, 160 cins ve 368 tür saptandı. Her iki grupta en sık saptanan filum Proteobacteria oldu. İki grup arasında filum, cins ve tür sayısı açısından anlamlı fark saptanmadı (p>0.005). 4 primer endodontik infeksiyon örneğinde Methanibrevibacter oralis ve Candidatus nitrosoarchaeum limnia arkeaları saptandı. Sekonder/persistan endodontik infeksiyonların mikrobiyal topluluklarının primer endodontik infeksiyonlar kadar çeşitli ve zengin olduğu gösterilmiştir. Primer endodontik infeksiyonlardan ilk defa Thaumarchaeota filumuna ait Candidatus nitrosoarchaeum limnia türü saptanmıştır.
