Kolorektal kanser hücrelerinde Siklooksijenaz -2 (COX-2)’nin sodyum bütirat aracılı transkripsiyon sonrası regülasyonu

Öz:
Kısa zincirli yag asitleri bagırsakta komensal bakteriler tarafından üretilmektedir ve bunların inflamasyonu azalttıkları bilinmektedir. Saptanan mekanizmalar içerisinde, siklooksigenaz-2 (COX-2) gibi inflamatuvar proteinlerin ve sitokinlerin ifadesinde transkripsiyonel inhibasyon bulunmaktadır. Birkaç çalısma bütiratın inflamatuvar genlerin mRNA kararlılıgını degistirebilecegini belirtmektedir; fakat mekanizmalar açıklanmamıstır. Bu projede, kolon kanseri hücre hatlarının sodyum bütirat ile muamele edilmesinin, birçok inflamatuar mRNA?nın kararlılıgını arttırdıgı bilinen bir RNA?ya baglanan protein olan HuR?nin sitoplazmadaki düzeyinde azalmaya neden oldugunu tespit edilmistir. Mekanistik olarak, sitoplazmik HuR miktarındaki azalmanın, HuR?un fosforlanmasını ve çekirdekte tutulmasını düzenleyen protein Chk2?nin fosforlanmasındaki artma ve p38 ile MK2 proteinlerinin fosforlanmasındaki azalma aracılıgıyla oldugunu gösterilmistir. Raportör analiz deneyleri ise HuR?nin azalan sitoplazmik seviyesinin COX-2 mRNA düzeyinde ve HuR baglanan sekansların 3?UTR aktivitesinde azalmaya neden oldugunu ortaya koymustur. Ek olarak yapılan RNA immün çöktürme deneyleri bütirat muamelesinin HuR?nin COX-2 3?UTR?sine baglanmasında önemli ölçüde azalmaya neden oldugunu göstermistir. Bu çalısmadan elde edilen veriler söyle özetlenebilir: bütirat sadece transkripsiyonel düzenleme ile degil aynı zamanda mRNA de-stabilizasyonu ile post-transkripsiyonel olarak da inflamatuvar genlerin ifadesini azaltabilmektedir.
