Leptin ve fizyopatolojik olaylardaki rolü

Öz:
Adiposit kaynaklı bir hormon olan leptin, beslenme ve enerji homeostazınde önemli bir rol oynamaktadır. Leptin yapıca sitokinlere olan benzerlikleri nedeniyle, sitokin olarak da sınıflandırılabilmektedir. Leptin eksikliği veya rezistans durumları insanlarda obezite, diyabet ve infertilite ile sonuçlanmaktadır. Leptinin antiobezite, üreme, hematopoez, anjiogenez, kan basıncı, büyüme, kemik hacmi, lenfoid organ homeostazı ve T lenfosit sistemleri gibi birçok sistemde temel etkileri geniş olarak gösterilmiştir. Leptinin büyümede de etkili olduğunu gösteren çalışmalar vardır. Büyüme hormonu tedavisinin vücut ağırlığını azaltarak dolaylı bir şekilde leptin düzeyini azalttığı gösterilmiştir. Leptin santral veya periferik karmaşık biyolojik etkilerini reseptörleri aracılığı ile düzenlemektedir. Leptin reseptörleri sınıf 1 sitokin reseptör superfamilyasındandır. Leptin reseptörleri santral sinir sistemindeki gibi, hematopoetik ve immun sistemler de dahil olmak üzere periferik dokularda yaygın olarak bulunmaktadırlar. Leptin reseptörlerinin en az 5 izoformu bulunmaktadır. Leptin reseptör izoformları çok çeşitli dokularda gösterilmiştir. Leptinin santral etkilerinin yanında bu reseptörlerin dokulardaki varlığı leptinin bu hücreler ve dokular üzerinde direkt etkisinin de olduğunu göstermektedir

Role of leptin on physiopathologic actions

Öz:
Leptin is an adipocyte-derived hormone that acts as a major regulator for food intake and energy homeostasis. Because of the similarities to cytokines, also identified as cytokine. Leptin deficiency or resistance can result in profound obesity, diabetes, and infertility in humans. Our understanding of leptin's biological functions has expanded from antiobesity to broad effects on reproduction, hematopoiesis, angiogenesis, blood pressure, growth, bone mass, lymphoid organ homeostasis, and T lymphocyte systems. Some studies show that leptin acts on growth. Growth hormone therapy reduce the level of leptin by decreasing body weight. Leptin orchestrates complex biological effects through its receptors, expressed both centrally and peripherally. Leptin receptor belongs to the class I cytokine receptor superfamily. The leptin receptor is expressed in the central nervous system, as well as in a wide spectrum of peripheral tissues, including the hematopoietic and immune systems. At least five isoforms of leptin receptor exist. Beside central effects the presence of leptin receptors in tissues throughout the body suggests that leptin can also have direct effects on cells and tissues.
