Rutin’in Deltamethrin ile Mide Toksisitesi Geliştirilen RatlardaOksidatif Stres, Apoptoz ve İnflamasyon ParametreleriÜzerine Etkisinin Araştırılması

Öz:
Deltamethrin (DLM) böceklerle ve çeşitli haşerelerle mücadelede yaygın olarak kullanılan oldukçaetkili bir insektisittir. DLM böceklerin kontrolünde kullanılırken, bilinçsiz kullanım sonucu ciddi toksiketkilere de sebep olabilmektedir. Rutin (RUT), turunçgillerden elde edilen ve düşük toksik etkilereve oksitlenmeme özelliğine sahip kuvvetli bir doğal antioksidan moleküldür. Bu çalışma ratlardaDLM kaynaklı oluşan mide toksisitesine karşı RUT’un etkilerinin araştırılması amacı ile planlandı.Deneysel işlem 28 gün olarak planlandı ve 5 grup oluşturuldu. Ratlara 28 gün boyunca oral yolla,DLM (1.28 mg/kg) veya RUT (25 ve 50 mg/kg) uygulandı. Mide dokusunda indirgenmiş glutatyon(GSH), lipit peroksidasyon ürünlerinden olan malondialdehit (MDA) seviyeleri ile süperoksitdismutaz (SOD), katalaz (KAT), glutatyon peroksidaz (GPx) aktiviteleri belirlendi. Tümör nekrozfaktörü alfa (TNF-α), nükleer faktör (eritroid kaynaklı 2)-related factor 2 (Nrf-2) ve nükleer faktörkappa B (NF-κB) gibi inflamasyon belirteçleri ile sistein aspartat spesifik proteaz kaspaz-3 (kaspaz 3) ve sinyal dönüştürücü ve transkripsiyon aktivatörü-3 (STAT3) apoptotik belirteçlerinin mRNAtranskript seviyeleri ölçüldü. Elde edilen sonuçlar doğrultusunda RUT uygulamasının DLM’ninneden olduğu oksidatif hasarı azalttığı, apoptoz ve inflamasyon belirteçleri üzerine düzenleyicietkileri olduğu tespit edildi.

Öz:
Deltamethrin (DLM) is a highly effective insecticide widely used to control insects and variouspests. While DLM is used to control insects, it can also cause serious toxic effects as a result ofunconscious use. Rutin (RUT) is a powerful natural antioxidant molecule obtained from citrus fruitsand has low toxic effects and non-oxidation properties. This study was designed to determine theprotective effects of RUT against gastric toxicity caused by DLM in rats. Rats were divided into 5groups and experimental procedure carried out for 28 days. DLM (1.28 mg/kg) and RUT (25 and 50mg/kg) were administered orally to animals. The levels of reduced glutathione (GSH),malondialdehyde (MDA), one of the lipid peroxidation products, and superoxide dismutase (SOD),catalase (CAT), glutathione peroxidase (GPx) activities were determined in the gastric tissue.mRNA transcript levels of inflammation markers such as tumor necrosis factor alpha (TNF-α),nuclear factor (erythroid-derived 2)-related factor 2 (Nrf-2) and nuclear factor kappa B (NF-kB) andapoptotic markers such as cysteine aspartate-specific protease caspase-3 (caspase-3) and ofsignal transducer and activator of transcription-3 (STAT3) were measured. According to our resultsit was determined that RUT administration reduced the oxidative damage caused by DLM and hadregulatory effects on apoptosis and inflammation markers.
